About the start-up
Gordian Biotechnology is a discovery stage biotechnology company based in South San Francisco. Gordian discovers treatments for age-related diseases using technology that lets us put hundreds of potential therapies into a single animal and independently measure the effects of each one. By reducing screening costs by ~100x, this unlocks more expensive, but more realistic animal models, such as spontaneous disease in species with human-like physiology.

Position/Project summary
Platform Visiting Student
Your mission as a platform visiting student is to assist in the creation and validation of gene therapies targeting complex diseases, generating the data we need to decide which therapies to move towards human trials. You’ll execute common and proprietary protocols within four laboratory focus areas: plasmid cloning and production, tissue dissociation, flow cytometry, and sequencing.
Main responsibilities will include (but are not limited to):
Assisting with the creation of gene therapy constructs.
Processing tissue from large animal models of disease into dissociated single cells
Running single cell RNA-seq assays
Communicating results with managers and leadership
The findings from this project will help guide Gordian to identify drug targets to reverse disease biology in chronic diseases such as osteoarthritis, NASH, heart failure, and idiopathic pulmonary fibrosis
Required skills/ Qualifications
Basic molecular biology techniques such as PCR
Strong preference for at least one of the qualifications in the section below
Preferred qualifications:
Plasmid cloning and production: standard DNA cloning techniques including PCR, gel electrophoresis, bacterial transformation, picking colonies, mini/midi-prep. Experience in plasmid design/maintenance on Benchling is a plus.
Sequencing: Creating and processing sequencing libraries. Experience with single cell technologies or Illumina sequencers is a plus.
Tissue dissociation: Taking tissues from our in vivo screens and turning them into single cell suspensions. Experience with animal sacrifices, fixation, and RNA quality control for RNA-seq are all pluses.
Flow cytometry: Ability to calibrate a cytometer, set gates to segregate cells from debris, and determine the percentage of fluorescent cells in a population. Experience with sorting and multiple fluorophores is a plus.
